CryoEM is a microscopy technique that applies a freeze-fixation technique to obtain the structure of biological macromolecules by using transmission electron microscopy to view samples at low temperatures. The sample is cryo-fixed and placed in an electron microscope where electrons are scattered in the sample and nearby ice. CD BioSciences uses a detector and lens system to image and record the scattered signals, which are then processed using 3D reconstruction techniques to obtain the 3D structure of the sample.
Types of sample holders available:
1. In-situ heating and biasing:Protochips Aduro 500 (link to in-situ page)
2. In-situ liquid and electrochemistry:Protochips Poseidon 510 (link to in-situ page)
3. Fischione tomography (+/- 70° tilt)
4. Cryo-holder and cryo-transfer holder for looking at frozen hydrated samples.
Typical Applications
1. Microstructure and nanostructure: size and morphology
2. Cross-section analysis (layer thickness, interface quality)
3. Crystal structure determination through electron diffraction
4. Defect analysis (dislocations, stacking faults, etc.)
5. High resolution images (~0.2 nm resolution)
6. Chemical information-composition and bonding (EDS, EELS) from single points, line scans or maps
7. Energy filtered imaging (EFTEM)
8. Z-contrast imaging using high angle annular dark field (HAADF)
Application Examples
1. Z-Contrast Imaging of InAs Quantum Wells in GaAs/AlAs Quantum Wells
2. Nanocrystalline Diamond in Ru-Doped DLC Films
3. XPS and HR-TEM Analysis of High k Thin Films
