CryoEM Imaging Service—FEI Mark IV Vitrobot

Cryo-electron microscopy (CryoEM) applies cryo-fixation technology to observe samples with a transmission electron microscope at low temperatures, to obtain the microscopic technology of the structure of biological macromolecules. After the piece is frozen and placed in the electron microscope, the electrons are scattered through the sample and the nearby ice layer. Use the detector and lens system to image and record the scattered signal, then process the signal, and finally use the three-dimensional reconstruction technology to obtain the three-dimensional structure of the sample. CryoEM imaging services are provided by CD BioSciences based on FEI Mark IV Vitrobot.

Introduction

1. Extremely consistent vitrified ice for frozen samples
2. Enclosed controlled environment with a working temperature of 4-60℃ (Peltier controlled) and able to maintain relative humidity at 100% with ultrasonic humidification
3. Small volume sample application with user defined software control of multiple sample applications, application time and wait time
4. Adjustable blotting parameters include the number of blot actions, blot duration, longitudinal grid positioning, and drain time between blotting and vitrification steps
5. Automated shutter control, grid and coolant container positioning

Summary

The FEI Mark IV is the fourth generation of sample vitrification robot from FEI. The robot controls environmental conditions, blot times, and plunge freezing to produce a consistent ice thickness for frozen vitrified samples.
