The spinning disk stereology setup is a fully motorized upright microscope from olympus equipped with the olympus disk spinning unit, a color camera and a very light sensitiv EM-CCD camera (color camera: QImaging Retiga 2000R, 1600 × 1200 pixels, 12 bit, linear full well 40,000 e, readout noise 16 e, QE 55%, 10 frames/sec @ 1600 × 1200 pixel; EM-CCD camera: Hamamatsu C9100-02, 1000 × 1000 pixels, 14 bit, quantum efficiency >
60%, full well capacity 70,000 e, readout noise < 1 e, 30.1 frames/sec @ 1000 × 1000 pixels). The Stereo Investigator software from micro bright field allows performing all common stereological procedures (i.e. optical disector or cavalieri) including widefield, epi-fluorescence and co-localization analysis. The setup can also be used for standard spinning disk or confocal imaging experiments.
