Cryo-electron microscopy (CryoEM) applies cryo-fixation technology to observe samples with a transmission electron microscope at low temperatures, to obtain the microscopic technology of the structure of biological macromolecules. After the piece is frozen and placed in the electron microscope, the electrons are scattered through the sample and the nearby ice layer. Use the detector and lens system to image and record the scattered signal, then process the signal, and finally use the three-dimensional reconstruction technology to obtain the three-dimensional structure of the sample. CryoEM imaging services are provided by CD BioSciences based on Leica Fluorescence CLEM.
This microscope allows the visualization of frozen cells, to determine, by the use of a fluorescent signal, the regions of interest where excess material has to be removed for high-resolution inmaging. The coordinates of the regions of interest can be saved and transfered to the other instruments down the pipeline, making for a considerably smoother workflow.
