Confocal microscopy is an optical imaging technique that uses point-by-point illumination and spatial pinhole modulation to remove scattered light from the non-focal plane of the sample, allowing for improved optical resolution and visual contrast compared to traditional imaging methods. The MMI CellCut/CellManipulator system is mounted on an Olympus IX-71 microscope with a motorized stage and CCD camera. Differential Interference Contrast and widefield fluorescence modalities are available. The optical tweezers use a 1064 YAG infrared laser with 3W trapping power that can be split (time-share mode) to trap up to 10 different objects. Dual quadrant detectors are available for detection.
Optical tweezers are capable of measuring picoNewton forces involved in single molecule dynamics and interactions (e.g. proteins & nucleic acids), while the laser microdissection setup can isolate tissue, single cells, or even submolecular structures (e.g. chromosomes) ; isolated material can then be prepared for genomic or proteomic analysis.
