Biological Imaging Service—Zeiss LSM 710 Confocal Microscope

Biological imaging is an important research tool for understanding tissue structure and elucidating the various physiological functions of living organisms. It uses light or electron microscopy to obtain direct images of the microstructure of biological cells and tissues, and the resulting images are analyzed to understand the various physiological processes of biological cells. Introduction

1. Laser lines
The LSM 710 is capable of 34 spectral detection channels using six laser lines-405, 458, 488, 514, 561 & 633 nm, plus an additional 594 nm HeNe laser (includes replacement main beamsplitter wheel equipped for 458, 488, 514, 561, 594 & 633 nm laser lines). The 561 and 594 lasers are needed to visualize mCherry, Texas Red, and other red fluorophores including Alexa 568, Alexa 594.
2. Fluorescence Equipment
The LSM 710 uses a six position turret that houses cubes holding a DAPI fluorescence filter, a Cy3 fluorescence filter and a GFP fluorescence filter.
3. Transmitted light equipment
The transmitted light source uses a halogen bulb for transmitted light source and a mirror to switch between transmitted light source and transmitted PMT (detector).
4. HBO Illuminator
The fluorescence light source uses a mercury bulb and power supply along with a touch pad for microscope control with docking station.
