Scanning Probe Microscope Imaging Service—Zeiss AxioObserver with 880-Quasar

The Scanning Probe Microscope (SPM) is an international surface analysis instrument developed in recent years, which is a high-tech product that integrates optical, mechanical, and electrical technology by using optoelectronics, laser technology, weak signal detection technology, applied optics technology, computer high-speed acquisition and control, and high-resolution graphics processing technology and other modern technological achievements. CD BioSciences offers Zeiss AxioObserver with 880-Quasar-based scanning probe microscopy imaging services.

Name of the Instrument/Platform

Zeiss AxioObserver with 880-Quasar

MANUFACTURER:
Zeiss

Introduction

Specifications
1. Zeiss AxioObserver with 880-Quasar (34-channel spectral, high-sensitivity gallium-arsenide phosphide detectors, GaAsP) confocal module & one-color fluorescence correlation spectroscopy (FCS)
2. Airyscan FAST super-resolution (max 2× better than confocal) with zoom optics (compatible with all objective magnification) and high-speed acquisition (max 4× faster, but less than maximal resolution)
3. Environmental chamber for 37℃, CO2, & humidity (35mm dishes or chamber slides)
4. Definite Focus II nosepiece (fast auto-focus to compensate for focus drift during sample heating)
5. NA 0.55 condenser (dry only) with DIC for 20×, 40×, 63× and 100× lenses
6. Objectives:
1) 10×/0.45 PlanApo (dry, no DIC)
2) 20×/0.80 PlanApo, with DIC (dry)
3) 40×/1.30 PlanNeofluar oil, with DIC
4) 40×/1.20 C-Apo water, with DIC, upon request
5) 63×/1.4 PlanApo oil, with DIC
6) 100× PlanApo oil, with DIC, upon request
7. 8 laser lines available:
1) 405 (diode laser),
2) 458, 477, 488, 5

Feature Highlights

Super-resolution (up to 2× better than standard confocal) for all objective magnifications with laser-scanning ease. Maximum resolution ~110 nm.
FAST module allows balancing speed with resolution (as much as 4× faster than standard Airyscan).

Applications

1. Single confocal images or image stacks for 3D viewing
2. FRAP with user-defined photobleach marks
3. CFP/YFP FRET
4. Photoactivation at 405 nm of caged dyes and other molecules
5. Colocalization of different fluorescent signals
6. Speed and versatility
7. Quasar "fingerprint" system separates very closely spaced fluorochromes. Example: DPPIV-GFP (Golgi, green) and Albumin-Alexa 488 (ER-Golgi, red)
