Confocal microscopy is an optical imaging technique that uses point-by-point illumination and spatial pinhole modulation to remove scattered light from the non-focal plane of the sample, allowing for improved optical resolution and visual contrast compared to traditional imaging methods. CD BioSciences, the imaging experts, can provide you with Nikon Ti-HCS Microscope-based confocal microscopy imaging services.
Objectives
1. Nikon Plan Fluor 4× NA 0.13 (working distance 16.5 mm)
2. Nikon Plan Fluor 10× NA 0.3 phase contrast (working distance 16mm; corrected for 0.17 mm coverslips (= type #1.5))
3. Nikon Plan Apo 20× NA 0.75 (working distance 1.0 mm)
4. Nikon S Plan Fluor ELWD 20× NA 0.45 (working distance 8.2 - 6.9 mm; adjustable for glas and plastic bottom dishes with a thickness between 0 and 2 mm)
5. Nikon Plan Apo 40× NA 0.95 (working distance 0.25 - 0.17 mm)
6. Nikon S Plan Fluor ELWD 40× NA 0.60 (working distance 3.6 - 2.8 mm; adjustable for glas and plastic bottom dishes with a thickness between 0 and 2 mm)
Camera
1. Nikon DS-Qi2 high-sensitive CMOS monochrome camera with 2404 × 2404 pixel resolution (in total 4908×3264, the image is cropped to fit the FOV of the microscope) and a quantum efficiency of 80% at 500 nm.
2. With the DS-Qi2 camera at 2404 × 2404 pixel resolution, the following pixel sizes result: 4× 1.83 µm; 10× 0.73 µm; 20× 0.37 µm; 40× 0.18 µm.

The Ti-HCS is a fully automated, inverted Nikon Ti2 microscope made for high content screening tasks. The microscope is encased by a environmental box from OkoLab for temperature-, CO2- and humidity control. By a software module names "JOBS" in the Nikon NIS-Elements acquisition software it is possible to screen in x/y, z and t everything from a LabTek dish to a multiwell plate. Images can be taken in brightfield and epifluorescence.
