Cryo-electron microscopy (CryoEM) applies cryo-fixation technology to observe samples with a transmission electron microscope at low temperatures, to obtain the microscopic technology of the structure of biological macromolecules. After the piece is frozen and placed in the electron microscope, the electrons are scattered through the sample and the nearby ice layer. Use the detector and lens system to image and record the scattered signal, then process the signal, and finally use the three-dimensional reconstruction technology to obtain the three-dimensional structure of the sample. CryoEM imaging services are provided by CD BioSciences based on Vitrobot Marc IV.
1. The process of plunging, blotting, and vitrification is fully automated upon placing a vial into the chamber and setting up the ethane holder.
2. Operation is strictly controlled by dedicated hardware and software, implying that parameters and results are reproducible.
3. This enables a high throughput of vitrified samples, with an easy and straightforward control of the vitrification process.
4. The parameters to be influenced are: temperature, humidity, the number of blots, and a number of critical time settings.
5. In addition to this automated plunger, our lab also has a uniquely crafted manual plunger for anyone who would rather perform this process manually.
The Vitrobot™ (Vitrification Robot) is a fully PC-controlled device for vitrification (rapid cooling) of aqueous samples.
